| R2R3-MYB转录因子家族是MYB转录因子中最大亚家族,在植物多种胁迫响应中扮演重要角色。为了筛选出与磷利用效率相关的R2R3-MYB成员,以最新组装的大麦MorexV3基因组对大麦中的R2R3-MYB转录因子家族进行了鉴定与生物信息学分析。结果表明,在大麦MorexV3基因组中鉴定出127个R2R3-MYB基因,并命名为Hv2RMYB-1~Hv2RMYB-127。这些基因分布于大麦7条染色体上,且启动子区富含激素响应顺式作用元件。经理化性质分析,大多数成员编码酸性且不稳定的蛋白质。通过系统发育及共线性分析将大麦R2R3-MYB家族划分为5个不同的分支及16对串联复制基因。 结合转录组数据(RNA-Seq)和实时荧光定量PCR技术(qRT-PCR),发现10个大麦R2R3-MYB基因在低磷胁迫下均有不同程度的差异性表达,其中Hv2RMYB-48显著上调表达。Hv2RMYB-48定位于细胞核,可以与MYB-ABRE-MYB顺式作用元件进行互作。 |

| The R2R3-MYB transcription factor family, the largest subfamily of MYB transcription factors, plays critical roles in various plant stress responses. However, its function under low-phosphate(P) stress remains poorly characterized. In this study, a total of 127 R2R3-MYB genes were identified in the barley MorexV3 genome using bioinformatic approaches and designated as Hv2RMYB-1 to Hv2RMYB-127. These genes were predominantly distributed on the 7 barley chromosomes and were found to harbor hormone-responsive cis-acting elements in their promoter regions. Physicochemical analysis revealed that most members encode acidic and unstable proteins. Phylogenetic and collinearity analyses classified the barley R2R3-MYB family into five distinct clades, including 16 pairs of tandemly duplicated genes. Based on transcriptomic data(RNA-Seq) and quantitative real-time PCR(qRT-PCR), 10 barley R2R3-MYB genes showed differential expression under low-Phosphorus stress, among which Hv2RMYB-48 was notably upregulated. Further investigation confirmed that Hv2RMYB-48 localizes to the nucleus and can bind to the MYB-ABRE-MYB cis-element. |
