      WRKY蛋白是一类转录因子,参与调控植物的多种生长发育和胁迫应答过程。为进一步探究WRKY在小麦响应逆境中的作用,本研究克隆了小麦中国春的TaWRKY72B基因,并对其进行了生物信息学、亚细胞定位和表达模式分析。结果表明,中国春的TaWRKY72B基因编码320个氨基酸,蛋白序列包含典型的WRKY保守结构域和C2H2锌指结构,分子量为33.94 kDa,理论等电点为6.60,是酸性亲水的不稳定蛋白,无信号肽序列,属非跨膜蛋白。二级结构预测表明,TaWRKY72B主要由无规则卷曲( 61.25%)、α-螺旋(23.12%)、延伸链(11.88%)和β-转角(3.75%)组成。TaWRKY72B定位于细胞核,符合转录因子特征。系统进化树分析显示,TaWRKY72B蛋白与节节麦、大麦和水稻等禾本科作物的WRKY亲缘关系较近。启动子顺式作用元件预测结果表明,该基因含有多个与生长发育、激素信号途径以及非生物胁迫应答相关的顺式调控元件。TaWRKY72B表达受激素、高温、低温和盐诱导,推测该基因可能参与小麦逆境胁迫应答和多种激素信号途径。

      WRKY proteins are a class of transcription factors that participate in the regulation of various plant growth, development and stress response processes. In order to further explore the function of WRKY in wheat response to adversity, TaWRKY72B gene was cloned, and bioinformatics, subcellular localization and expression pattern analysis were carried out. The results showed that TaWRKY72B gene encoded 320 amino acids, and the protein sequence contained the typical WRKY conservative domain and C2H2 zinc finger structure. The molecular weight was 33.94 kDa, and the theoretical isoelectric point was 6.60. It was an acidic hydrophilic unstable protein, without signal peptide sequence, and belonged to non-transmembrane protein. The secondary structure prediction showed that TaWRKY72B was mainly composed of irregular curls(61.25%), α-spiral(23.12%), extended chain (11.88%) and β-corner(3.75%). TaWRKY72B was located in the nucleus, consistent with the characteristics of transcription factors. The phylogenetic tree analysis showed that TaWRKY72B protein was closely related to WRKY of gramineous crops such as Aegilops tauschii, Hordeum vulgare and Oryza sativa. The prediction of cis-acting elements in the promoter showed that the gene contained many cis-acting elements related to growth, development, hormone signaling pathway and abiotic stress response. TaWRKY72B is induced by hormones, high temperature, low temperature and salt, and thus it is speculated that this gene may participate in the response of wheat to stress and multiple hormones signaling pathways.
