      为明确拔节期低温对小麦新品种漯麦163的生理生化以及抗寒基因表达特性的影响,以漯麦163及其母本漯麦6010为材料,于拔节期在人工智能低温室进行低温胁迫,分析低温胁迫对两品种叶片叶绿素含量、超氧化物歧化酶(SOD)活性和脯氨酸含量的影响,并对抗寒相关基因(SODWCS120LEAP5CS)的表达模式进行分析。结果表明,持续低温胁迫下,两个品种的叶绿素含量均显著下降,SOD活性和脯氨酸含量均呈现先升后降的变化趋势。对抗寒相关基因的表达模式进行分析,发现与对照(低温处理0 d)相比,低温胁迫下,SODWCS120基因的表达量在两品种中于处理后不同时间点均显著下降;LEA基因的表达量在漯麦6010中于处理后不同时间点显著下降,而在漯麦163中于低温处理1 d后显著提高;P5CS基因的表达量在漯麦163于低温处理3 d后以及在漯麦6010于低温处理2 d和3 d后均显著提高。结合大田观察,发现漯麦163和漯麦6010均具有较强的耐倒春寒能力,可作为抗寒性亲本种质资源。

      In order to understand the effects of low temperature on the physiological, biochemical and cold-resistant gene expression characteristics of a new wheat variety Luomai 163 at jointing stage, Luomai 163 and its female parent Luomai 6010 were used as materials. Low temperature stress was applied in artificial climate chamber, and the effects of low temperature treatment on chlorophyll content, superoxide dismutase(SOD) activity and proline content of leaves, and the expression patterns of cold-resistance related genes(SOD, WCS120, LEA and P5CS) were analyzed. The results showed that under continuous low temperature treatment, chlorophyll content of the two varieties decreased significantly; SOD activity and proline content increased first and then decreased. The expression patterns of cold-resistance related genes were analyzed, and compared with the control(low temperature treatment for 0 d);the expression levels of SOD and WCS120 in the two varieties were decreased significantly at different treatment time. The expression levels of LEA gene were decreased significantly at different treatment time in Luomai 6010, but increased significantly in Luomai 163 after low temperature treatment for 1 d. The expression of P5CS gene was increased significantly in Luomai 163 after low temperature treatment for 3 d and in Luomai 6010 after low temperature treatment for 2 d and 3 d. Combined with field observation, Luomai 163 and Luomai 6010 showed strong cold tolerance, which can be used as a good germplasm resource for cold resistance, so as to speed up the breeding of cold resistant wheat varieties.
