      为探索低温胁迫下油菜素内酯(BR)对冬小麦分蘖节及叶片 wcor413-like基因表达的影响,以寒地冬小麦东农冬麦1号为试验材料,利用RT-PCR法克隆 wcor413-like基因的cDNA序列,用生物信息学法分析该序列的特征,用实时荧光定量PCR(qRT-PCR)技术检测5 ℃、0 ℃、-10 ℃和-25 ℃四个温度处理下分蘖节和叶片中 wcor413-like基因的表达量。结果表明, wcor413-like基因含有一个629 bp的开放阅读框,可编码209个氨基酸,编码的wcor413-like蛋白为稳定蛋白质,主要分布在细胞质中。qRT-PCR分析表明,在叶片中, wcor413-like基因的表达量随着温度的降低呈现先上升后下降的变化趋势,-10 ℃时表达量最高;在分蘖节中, wcor413-like基因的表达量随着温度的降低呈持续上升的趋势,-25 ℃时表达量最高。BR处理后的叶片和分蘖节中, wcor413-like基因的表达量均在-25 ℃时达到最高,叶片中该基因的表达量显著高于对照组,而分蘖节中该基因的表达量显著低于对照组。推测BR可通过调控叶片和分蘖节中 wcor413-like基因的表达来提高冬小麦叶片的抗寒性。

      In order to explore the effect of brassinosteroids (BR) on the expression of wcor413-like gene in tiller nodes and leaves of winter wheat under low temperature treatment, the cold winter wheat Dongnongdongmai 1 was used as the experimental material, and wcor413-like was cloned by RT-PCR method. The cDNA sequence of the wcor413-like gene was analyzed by bioinformatics, and the real-time quantitative PCR (qRT-PCR) method was used to determine the expression of wcor413-like gene in tillers and leaves at four temperatures (5 ℃, 0 ℃, -10 ℃ and -25 ℃) after BR treatment. The results showed that wcor413-like gene contains a 629 bp open reading frame, encoding 209 amino acids. wcor413-like protein is a stable protein, and mainly localizes in the cytoplasm. qRT-PCR analysis showed that with the decrease of temperature, the expression level of wcor413-like gene in leaves increased first and then decreased and reached to the maximum at -10 ℃; The expression of wcor413-like gene in tiller nodes increased continuously, and the maximum appeared at -25 ℃. After BR treatment, the expression of wcor413-like gene in leaves and tiller nodes reached the highest level at -25 ℃, and the expression of wcor413-like gene in leaves were significantly higher than that in control group, while the expression of wcor413-like gene in tiller nodes were significantly lower than that in control group. It was speculated that BR can improve the cold resistance of winter wheat by regulating the expression of wcor413-like gene in leaves and tiller nodes.
