      Vromindolines(Vins)是一种燕麦属(Avena L.)特异的淀粉结合蛋白,能够与淀粉体膜脂相互作用,影响籽粒硬度。为了解 Vin-2基因与裸燕麦籽粒硬度的关系,选用裸燕麦核心种质材料街棚莜麦对燕麦 Vin-2 AnVin-2)基因进行克隆和等位变异检测,发掘 AnVin-2基因等位变异的检测标记,探究 AnVin-2A AnVin-2C AnVin-2D基因在籽粒不同发育时期以及4种不同硬度裸燕麦材料(2份软质,2份硬质)成熟籽粒中的表达模式;并选择10份不同硬度裸燕麦材料(5份软质,5份硬质)对 AnVin-2C4的等位变异进行鉴定。结果表明, AnVin-2基因在裸燕麦中存在14种等位变异,分别分布在4A、7C和4D染色体上,均无内含子,包含完整的开放阅读框444 bp,可编码147个氨基酸;根据序列差异开发出 AnVin-2基因在A、C和D染色体组上的标记以及 AnVin-2C1 AnVin-2C4基因的特异标记,且 AnVin-2C4在10份不同硬度材料中存在5种等位变异。表达分析发现, AnVin-2C AnVin-2D基因在籽粒不同发育时期的表达量总体均呈先升后降的变化趋势,但在4份不同硬度材料成熟籽粒中没有明显差异(除冀杂二号成熟籽粒中 AnVin-2C基因的表达量显著低于73014-336、8399-3-4和7929-1-6成熟籽粒中的表达量外), AnVin-2A在籽粒不同发育时期均没有检测到表达。以上结果表明,裸燕麦 AnVin-2基因以多拷贝形式存在,但其序列多态性与籽粒硬度没有明显关联。

      Vromindolines (Vins) are starch-binding proteins of Avena genus, which can affect the grain hardness by interacting with amyloid lipids. In order to find out the relationship between AnVin-2 gene and oat grain hardness, ‘Jiepengyoumai’ was selected for cloning oat Vin-2 ( AnVin-2 ) gene and detecting the allelic variations. The AnVin-2 gene markers of A, C and D subgenomes of oat were developed, and the expression patterns of AnVin-2A, AnVin-2C and AnVin-2D at different grain developmental stages in 4 mature grains of oat materials (2 soft and 2 hard) were studied. The relationship between AnVin-2C4 and grain hardness were identified by comparing the allelic variation of AnVin-2C4 in 10 different hardness materials(5 soft and 5 hard). Results showed that 14 allelic variations of AnVin-2 gene in oat were cloned, which were mapped on chromosomes 4A, 7C and 4D. All the 14 sequences contain a complete 444 bp open reading frame, encoding 147 amino acids without introns. The genomic markers of AnVin-2 on A, C, and D were developed based on sequence differences among chromosomes. Two markers were developed, which can be used to specifically amplify AnVin-2C1 and AnVin-2C4, and five allelic variations of AnVin-2C4 were identified in 10 materials. Analysis of the expression patterns of AnVin-2C and AnVin-2D showed that the transcript levels of the two genes increased first and then decreased at seven growth stages of grain, but there was no significant difference among the four mature grains with different grain hardness(except the expression of Anvin-2C in Jiza 2 was significantly lower than other materials). However, expression of AnVin-2A was not detected at all developmental stages. In a word, the AnVin-2 genes existed in the form of multiple copies in naked oat, but its sequence polymorphisms were not significantly associated with grain hardness.
