为研究过氧化物酶基因 TaPRX-2A在调控小麦穗部性状中的功能,以苏麦3号为材料,通过Ensemblplants数据库获得 TaPRX-2A全长cDNA序列,利用PCR方法克隆获得该基因编码区全长序列。生物信息学分析结果表明,该基因包含一个1 026 bp的开放阅读框,编码342个氨基酸。进一步构建基因过表达载体 TaPRX-2A-PC186,通过基因枪介导的遗传转化方法将 TaPRX-2A-PC186转入小麦KN199中,创建过表达 TaPRX-2A转基因株系,调查 TaPRX-2A基因对转基因株系穗部性状的影响。结果表明,过表达 TaPRX-2A转基因株系的穗长显著变短,穗粒数显著减少,说明 TaPRX-2A在调控小麦穗部发育过程中具有重要的作用。 |

In order to study the function of peroxidase gene TaPRX-2A in regulating the spike traits of wheat, in this study, the cDNA sequence of TaPRX-2A was obtained from Ensembl plants database, and cloned by PCR method from the wheat variety Sumai 3.Bioinformatics analysis results showed that TaPRX-2A contained a 1 026 bp ORF (open reading frame), encoding a total of 342 amino acids. The recombinant expression vector TaPRX-2A-PC186 was further constructed, and then transformed into KN199, to create TaPRX-2A transgenic overexpression lines. The effect of TaPRX-2A on the spike traits length of transgenic lines was studied. The results showed transgenic plants overexpressing TaPRX-2A had shorter spike length and fewer kernel number per spike, suggesting that TaPRX-2A played an important role in regulating the development of wheat spike. |
