| 为进一步对类病斑突变基因 lm3进行克隆和功能研究,以EMS诱变普通小麦花培品系H261所得后代中选育出的一个稳定遗传的小麦类病斑突变体LF2010为研究材料,通过BSA(bulked segregant analysis)、MBSA(multiple bulked segregant analysis)、小麦90K基因芯片结合BSA三种不同的基因定位策略对该突变基因进行定位。结果表明, lm3基因位于小麦6B染色体的长臂,与其两侧标记SWES2和Xbarc134的遗传距离分别为13.1 cM和3.8 cM,为一个新的小麦类病斑突变基因。 |

| In order to clone the lesion mimic mutant gene lm3 and analyse its function further,a lesion mimic wheat mutant LF2010,obtained from an anther culture line H261 which was mutated with EMS,was chosen as material,and three kinds of different gene mapping strategies were taken to map it,including BSA(bulked segregant analysis),MBSA(multiple bulked segregant analysis),and wheat 90K gene chip combined with BSA.The result showed that a genetic linkage map containing 15 pairs markers linked with lm3 was obtained. Finally, lm3 was mapped on 6BL of wheat chromosome,13.8 cM away from marker SWES2 and 3.8 cM away from Xbarc134. lm3 is a new gene of wheat lesion mimic mutant. |
