      为探讨大豆DNA导入对春小麦农艺和品质性状的影响,在2012-2014年间,对宁春4号、宁春44号、高代品系J058号及其通过花粉管通道法分别导入大豆品种铁丰31号、承豆6号、汾豆78号和铁9808号基因组DNA的后代株系进行了籽粒蛋白质含量、湿面筋含量和沉降值及株高、穗长、穗粒数、穗粒重和千粒重等农艺性状分析,并利用RAPD标记对导入系进行了验证。结果表明,大豆基因组DNA导入系的农艺性状和品质性状与导入受体相比发生了明显的变化。以宁春4号为受体、铁9808号为供体的导入系019的经济系数和千粒重分别高出亲本14.33%和14.84%,且与受体差异显著。以宁春4号为受体、承豆6号为供体的导入系013在2012年所有导入系中蛋白质含量、湿面筋含量、沉降值均最大,两年试验的多重比较结果中仅导入系013的蛋白质含量与亲本宁春4号间差异显著,且较亲本高出13.33%。18个RAPD引物中有3个引物扩增出清晰带型,在受体小麦和供体大豆亲本间存在多态性,且部分导入系出现供体条带,如导入系013、015、019和020。试验最终获得了农艺性状优良、蛋白质含量增高的导入系013作为后续研究的重点。

      For a better understanding of common wheat quality breeding,three spring wheat varieties/lines,Ningchun 4,Ningchun 44,and J058,and their introgression lines created by pollen-tube pathway using genomic DNA of soybean varieties Tiefeng 31,Chengdou 6,Fendou 78 and Tie 9808,respectively,were used.Quality traits,such as protein content,wet gluten content and sedimentation were analyzed and plant height,spike length,thousand-kernel weight,grains per spike and other agronomic traits were also investigated after harvest at the mature period during 2012-2014.Besides,RAPD marker analysis was conducted.The results showed that related agronomic traits and quality traits of introgression lines had changed in comparison with parents.The economic coefficient and the thousand-kernel weight of introgression line 019 were significantly higher than those of the parent Ningchun 4 with an increase rate of 14.33% and 14.84%,respectively.In 2012,quality traits of introgression line 013 were reached highest level in all tested lines.Multiple comparison results revealed that only the protein content of introgression line 013 reached significant level,with the increase rate of 13.33%,compared to the parent Ningchun 4.Eighteen random primers were employed to amplify genomic DNA of the donor soybeans and the receptor wheat materials as well as their introgression lines.Of them,three RAPD markers gave clear bands to separate the soybeans from wheat evidently.Polymorphic bands were detected between wheat parents and soybeans as well as part of introgression lines which had donor bands,such as introgression lines 013,015,019 and 020.The introgression line 013 which had remarkable agronomic traits and improved protein content was finally selected as a new material for future study.
