      小麦籽粒硬度是其最为重要的品质性状之一,现已发现控制小麦籽粒硬度的主效基因是PinaPinb。为了丰富籽粒硬度新型等位基因,根据已报道的小麦PinaPinb基因的保守序列,设计了2对特异性引物,对粗山羊草(Aegilops tauschii)进行PinaPinb基因的PCR扩增、克隆及序列分析。结果表明,本研究共得到2个新型Pina等位基因和3个新型Pinb等位基因,核酸序列长度均为447 bp,编码148个氨基酸残基,具有麦类作物PINA和PINB蛋白所特有的色氨酸结构域:WRWWKWWK和WPTKWWK。与已知的PinaPinb基因序列相比较,这些基因含有多个SNP变异位点,丰富了小麦籽粒硬度改良的遗传资源。[JP]

      Grain hardness is one of the main quality characters of wheat. It has been revealed that the major genes controlling grain hardness in wheat were Pina and Pinb.According to the conserved regions of Pina and Pinb genes reported in wheat, two pairs of specific primers were designed and used to amplify in wheat D-genome progenitor, Aegilops tauschii.The results showed that two novel Pina alleles and three Pinb alleles were obtained. Sequence analysis indicated that all of two Pina genes and three Pinb genes were 447 bp in size,encoding 148 amino acid residues containing the tryptophan motif of WRWWKWWK and WPTKWWK, which were specific to PINA and PINB in cereal crop.Compared with the reported Pina and Pinb sequences, some SNPs were also identified in the Pina and Pinb alleles of Ae.tauschii. The result enriched genetic resources for kernel hardness improvement of wheat.
