      非生物逆境是小麦生长、发育及产量形成的主要限制因子之一。为揭示小麦抗逆胁迫响应机制,并为培育转基因抗逆种质奠定基础,本研究利用同源克隆的方法从普通六倍体小麦中国春中克隆出TaSKIP基因并进行序列分析及基因定位,同时对TaSKIP基因在PEG6000胁迫下的表达模式进行分析。结果表明,共克隆得到3个TaSKIP基因,其中2个基因被定位于6A和6D染色体上。序列分析表明,TaSKIP 6A编码区长为1 827 bp,编码608个氨基酸,TaSKIP BTaSKIP 6D编码区长均为1 824 bp,编码607个氨基酸。氨基酸序列比对分析表明,小麦TaSKIP蛋白与水稻、玉米和大麦等禾本科植物SKIP蛋白的同源性均大于88%,而且含有相同的 SKIP/SNW结构域。实时荧光定量PCR(Quantitative real time PCR,qRT PCR)检测结果表明,TaSKIP在模拟干旱胁迫条件下表达量上调,暗示该基因在小麦非生物逆境胁迫响应中起重要作用。

      Abiotic stress is one of the main limiting factors in wheat growth,development and yield.In order to lay a foundation for research of revealing the mechanism of wheat in response to stress and breeding the stress resistant germplasm,the homologous cloning strategy was used to clone TaSKIP gene from common hexaploid wheat cultivar Chinese Spring and quantitative real time PCR (qRT PCR) was used to analyse the expression patterns under the stress of PEG6000.The results showed three TaSKIP genes were obtained.And two were localized on chromosome 6A and 6D by using Chinese Spring nulli tetrasomes,respectively.Sequence analysis showed that the coding region of TaSKIP 6A is 1 824 bp long which encodes 608 amino acids,and the coding region of both TaSKIP B and TaSKIP 6D are 1 824 bp long which encode 607 amino acids.Multiple comparison analysis of amino acids showed that the homology of protein TaSKIP among wheat and Poaceae crops,such as rice,maize and barley is more than 88%,and all these TaSKIP proteins shared the identical SKIP/SNW motif.The qRT PCR results showed that the expression level of TaSKIP increased under the condition of drought stress in Chinese Spring,suggesting that TaSKIP plays an important role in response to abiotic stress in wheat.
