      2C型蛋白磷酸酶 (PP2C) 是ABA信号途径的关键组分,在ABA信号转导及植物对非生物逆境胁迫的应答过程中发挥着重要作用。为给 TaPP2C59基因功能的研究奠定基础,同时为小麦对非生物胁迫应答的信号转导机理研究提供参考,本研究从小麦中克隆了第一个PP2C基因 TaPP2C59,并对其在逆境胁迫以及多种信号分子处理下的表达模式进行研究。序列分析表明,该基因开放阅读框 (ORF) 为855 bp,编码284个氨基酸。进化树分析表明,该基因编码的蛋白与水稻 OsPP2C45蛋白和拟南芥AtPP2C59蛋白的亲缘关系最近。多序列比对分析表明, TaPP2C59具有PP2C家族蛋白的保守结构特征。实时荧光定量PCR分析表明,该基因的表达显著受ABA、低温和高盐胁迫抑制。因此,可以推测 TaPP2C59可能作为负调控因子参与ABA信号及非生物逆境胁迫应答。

      Protein phosphatase 2C (PP2C) genes are major components of ABA signaling, which plays an important role in the response of ABA signaling transduction and abiotic stresses. In order to lay a basis for the study on TaPP2C59 function and provide reference for the study on the mechanism involved in abiotic stress response in wheat, we isolated a PP2C gene designated as TaPP2C59 from wheat and investigated its expression patterns under various abiotic stresses and signaling molecules treatments. TaPP2C59 ORF was 855 bp, which encoded 284 amino acids. Phylogenetic analysis indicated that TaPP2C59 was closely related with OsPP2C45 and AtPP2C59. Comparison of TaPP2C59 with other known PP2C proteins suggested that TaPP2C59 had the conserved motifs of PP2C family. Real-time quantitative PCR (QPCR) assay revealed that the expression of TaPP2C59 was inhibited after ABA, low temperature and H2O2 treatments. Thus, it was concluded that TaPP2C59 might act as a negative factor in the response of ABA signaling and abiotic stresses.
