      为了给小麦远缘亲本的研究和利用提供参考,根据已定位于普通小麦1DS和1BL上的EST序列设计96对STS引物,对中国春、簇毛麦 、中国春 簇毛麦的1V附加系和易位系进行多态性分析,开发出4个簇毛麦1V染色体臂的STS标记。其中,BE499250 STS和BE591682 STS/RsaⅠ为共显性标记,可以扩增出1条1VS片段,同时还可以扩增出1条1DS片断,能将1DS和1AS、1BS 区分开来;BE581358 STS/HaeⅢ和BE585781 STS/RsaⅠ是显性标记,能分别扩增出2条和1条1VL片段,但扩增的小麦染色体片段不能区分1DL和1A、1B。这些标记已成功地应用于小麦 簇毛麦整臂互补易位系T1DL·1V#3S和T1DSo1V·3L的筛选。

      It is very important to develop the most reliable and informative molecular markers to expedite the screening of large numbers of progeny for wheat relative species translocations. In the present study, in order to develop STS markers for 1V chromosome arms of Haynaldia villosa, 96 pairs of STS primers were designed based on EST sequences downloaded from Graingenes mapped on wheat chromosome 1DS and 1BL using software primer 3.0, four STS markers were developed after testing those primers among Chinese Spring, Haynaldia villosa and translocation lines and additional lines derived from Chinese Spring and Haynaldia villosa. Two co dominant marker BE499250 STS and BE591682 STS/RsaⅠfrom 1DS could trace a polymorphic segment of 1VS of Haynaldia villosa, and a fragment of wheat 1DS which distinguished from 1A and 1B of wheat. The two dominant marker BE518358 STS / HaeⅢ and BE585781 STS/RsaⅠfrom 1BL could amplify two 1VL fragments and one 1VL fragment of Haynaldia villosa, respectively, and a wheat fragment which could not differentiate the 1D fragment with 1A and 1B. These four STS markers could be used to screen large progeny to identify individuals with 1VS or 1VL for two new compensating Robertsonian translocations T1DLo1V#3S and T1DSo1V#3L.
